A ready-to-use slide culture system for the cultivation of fungi. Simplify your fungal slide cultures while saving money, reducing frustration, and eliminating preparation time. Desiccation is prevented with a built in humidity chamber and the culture can be incubated for long periods of time. Device fits conveniently on the microscope stage for easy observation without disturbing the fungal structures. Transcript: The purpose of this demonstration is to show you how easy it is to prepare and examine a fungal slide culture using the MycoVue system Remove the lid from the MycoVue devise. Carefully peal off the protective tab that covers the culture media. Discard the tab. Inoculate the agar surface with your specimen. Place the included sterile cover slip on to the inoculated agar surface. Add approximately 1 ml of sterile water to the white sponge. This will keep the media hydrated and prevent shrinkage during incubation. Replace the lid of the device and incubate at 25 ºC. To examine growth, remove the cover and place the entire unit on the microscope stage. Examine he culture at 400 power which is the hi _____ objective lens. The MycoVue can be examined as often as you like without disturbing the delicate fungal structures. Alternatively, you may stain the fungal growth adhering to the cover slip. Do this by placing a drop of Lactophenol Cotton Blue or Hardy’s Blue Mount on a glass microscope slide. The remove the MycoVue cover slip and place it on the slide for viewing under the microscope. Replace the MycoVue cover slip with a new one. Traditional methods present some problems: 1. First of all, too much growth is removed for examination, making it difficult to view individual structures. Teasing apart the tangled mass is difficult. 2. The material is often taken from non-sporulating areas of growth. 3. The disruption of the fruiting structures may destroy the delicate arrangement of the spores and their supporting structures. 4. It is difficult to now the optimal time when the fruiting structures are mature enough to be removed for examination. 5.
