Do we not label the agar plate prior to inoculation? What culture was being used?

There may be several reasons why you want a single colony. One could be for describing the microorganism at a macroscopic level. Imagine if you had no isolated colonies, how well would you describe the microbe? It would be more difficult as the characteristic is jumbled up. And also, as you said, isolated colonies provide a way of making relatively sure that you don't have contaminents. Imagine having a lawn of microbes on the agar, could you easily distinguish the contaminent?

I jus have a question... Wy we need to sterilise the innocultaion loop very often, even while streaking every lines, actually we have been tought to sterilise ur loop only once ..that is before taking the organism and after completing ur total streaking in the agar plate..!!

Why do you want only a single colony to be isolated? Is this how you are sure it is the species you want and not a contaminant (because it came from one cell), or...?
