Why can't you just inoculate the slant straight from the bottle of beer?

You can usually incubate from 60F to 80F. It's not that important. Usually just leaving it at room temp. away from light is enough. Yeast on dishes doesn't last long as the thin layer of media will evaporate quickly. Slants can last months in a refrigerator (until they start turning gray and dying).

Could you give some more info on how to incubate the slants and dishes and on how long the yeast colonies can survive? Thanks

Over the top, I my opinion. This method is fine if your separating strains of yeast. Make a batch of wort pour the beer sediment in and let it reproduce. You will have a large yeast culture in 1 to 3 days.

Nice tutorial man. Now I can apply all I learned in Microbio to beer making. Yeast is expensive. I assume you could use any agar? What phase of growth are they being frozen in in the fridge?
